CD 34-Positive Cells and CFU-C Assay with GM-CSF in Human Bone Marrow, Peripheral Blood and Cord Blood

Abstract
Hematopoietic stem cell grafting can be done with cells from different origin; mononuclear cells (MNCs) from bone marrow (BM), peripheral blood (PB) and umbilical cord blood (CB) have been used as grafting stem cells and progenitor cells. CD34
monoclonal antibodies recognize a membrane surface phophoglycoprotein, selectively expressed within the lymphohematopoietic system on progenitors and stem cells.
We studied CD34-positive cell population and CFU-C (Colony Forming Unit in Culture) assay in BM, PB and CB MNCs in bone marrow from 10 healthy volunteers, umbilical cord blood from 20 babies at birth and peripheral blood from 10 patients during
recovery
phase following chemotherapy. were fractionated by ficoll gradient separation Anti-CD34 monoclonal antibody/flowcytometry technology was used for identification of hematopoietic stem cells. MNCs from BM, CB and PB were cultured in semisolid agar
media
for CFUc with and without Granulocyte Monocyte-Colony Stimulating Factor (GM-CSF).
Our study shows the difference of CD34-positive population in all three products, averaging in PB;1.09%, CB; 1.86%, BM; 2.61%and average number of CFUc with/without GM-CSF are in PB; 16, 8 1, 0 CB; 104.7/5.0, BM; 299.5 14.2 In this study MNCs
from BM,
CB and PB at lower rates, CFUc number was correlated with CD34-positive cell population, MNCs from cord blood and peripheral blood can be used as a source of autotransplanation.
